The values represent the average 50% inhibitory concentration (IC50) [ng/mL] (fold change relative to the reference virus). The reference viruses were RSV-A and RSV-B strains without AA substitutions. Neutralization assays were conducted to evaluate the changes in susceptibility to palivizumab, suptavumab, and nirsevimab-mAbs that specifically bind to sites II, V, and Ø of the F protein, respectively-due to amino acid (AA) variations. The assays included one or two strains of RSV with specific AA substitutions, as well as reference strains of RSV-A and RSV-B without any AA substitutions. RSV replication was measured using enzyme-linked immunosorbent assay. Subsequently, the IC50 values were calculated using a nonlinear fit algorithm. Each measurement was conducted in duplicate.

Fig 1. Epitope-specific antibodies (ESA) levels at sites Ⅱ, Ⅲ, Ⅳ, Ⅴ, and Ø Serum ESA levels, measured using competitive enzyme-linked immunosorbent assay (ELISA) [μg/mL], a re shown for antigenic sites Ⅱ (palivizumab), Ⅲ (MPE8), Ⅳ (clesrovimab), Ⅴ (suptavumab), and Ø (nirsevimab). Serum samples were collected from 92 healthy adults aged 28-75 yea rs (median, 41.5 yea rs) between Februa ry and Ma rch 2020. Significant differences were observed across all combinations of ESA concentrations (p<0.001 for a ll compa risons).
