Overview

Corynebacterium diphtheriae is a Gram-positive bacterium and the causative agent of diphtheria. Toxigenic strains can carry the tox gene through lysogenic corynephages, enabling production of diphtheria toxin (DT), the major virulence factor associated with disease development.

Research on C. diphtheriae mainly focuses on diphtheria toxin structure, toxin-host interactions, antibody recognition, and immune response analysis. Diphtheria toxin and its non-toxic variant CRM197 are important research targets for toxin detection, antibody development, and vaccine-related studies.

The structural and functional properties of DT make it an important model for studying bacterial toxins, antigen recognition, and antibody-mediated neutralization.

Diphtheria Toxin Structure

Diphtheria toxin (DT) is an exotoxin produced by toxigenic C. diphtheriae. The toxin consists of two functional regions: A fragment (catalytic domain), responsible for enzymatic activity and inhibition of host protein synthesis; and B fragment (binding/translocation domain), which mediates receptor binding and facilitates toxin entry into host cells. After entering host cells, the catalytic domain induces ADP-ribosylation of elongation factor-2 (EF-2), resulting in inhibition of protein synthesis and cellular dysfunction.
