Overview

Chikungunya virus (CHIKV) is an enveloped, mosquito-borne virus belonging to the genus Alphavirus in the family Togaviridae. It is primarily transmitted by Aedes aegypti and Aedes albopictus mosquitoes and causes chikungunya fever.

CHIKV infection is commonly associated with fever, rash, and pronounced joint and muscle pain. Research on CHIKV focuses on viral entry and replication, host-virus interactions, immune responses, antigenicity, and antibody-mediated neutralization.

The CHIKV genome is approximately 11.8 kb and contains two major open reading frames. The 5' ORF encodes the four non-structural proteins nsP1-nsP4, while the 3' ORF encodes the structural proteins Capsid, E3, E2, 6K, and E1.

CHIKV Structure

CHIKV is an enveloped virus approximately 60-70 nm in diameter. Its positive-sense single-stranded RNA genome is enclosed within a capsid and surrounded by a lipid envelope. The viral surface contains spike complexes formed by the E1 and E2 glycoproteins. Together, these proteins provide a framework for studying different stages of the CHIKV life cycle, from viral entry and membrane fusion to RNA replication and virion assembly.

E1 and E2 Glycoproteins Key Target
E1 and E2 form the major spike complexes on the CHIKV surface and are central to viral entry. E2 is involved in interactions with host cells and is an important target for antigen characterization, antibody recognition, and neutralization studies. E1 functions primarily as the viral membrane fusion protein and is therefore widely studied in research on viral entry, membrane fusion, and neutralizing antibodies. Because both proteins are exposed on the viral surface, E1 and E2 are also important targets for vaccine and immunogen research.

Capsid Protein Assembly
The Capsid protein associates with viral RNA to form the nucleocapsid and contributes to genome packaging and virion assembly. Capsid is therefore useful for research into viral structure, genome packaging, assembly, and antigenicity.

nsP1-nsP4 Replication
The four non-structural proteins nsP1, nsP2, nsP3, and nsP4 constitute the core CHIKV replication machinery. nsP1 is involved in membrane association and RNA capping, while nsP2 contributes to polyprotein processing and has helicase and NTPase activities. nsP3 participates in replication-complex formation and host-factor interactions. nsP4 is the viral RNA-dependent RNA polymerase (RdRp) responsible for viral RNA synthesis. These proteins are important targets for studies of viral replication, host-virus interactions, pathogenesis, and antiviral strategies.
