Overview of ASFV

African Swine Fever Virus (ASFV) is a large, enveloped, double-stranded DNA virus belonging to the family Asfarviridae. It causes African swine fever (ASF), a highly contagious and often fatal disease affecting domestic pigs and wild suids. ASFV infection can result in severe economic losses and remains a major challenge for the global swine industry.

ASFV has a large linear DNA genome that encodes a broad range of structural and non-structural proteins involved in viral entry, replication, assembly, host interaction, and immune evasion. Due to its complex biology and extensive immune evasion mechanisms, ASFV research focuses on multiple viral proteins rather than a single dominant target.

This complexity is reflected in the wide range of ASFV antigens studied, including p72, p54, p30, p17, and CD2v (EP402R), each playing distinct roles in viral infection and host interaction.

ASFV Structure

ASFV is characterized by a complex multilayered virion structure. From the outside inward, the mature virion contains a lipid envelope, outer membrane, capsid, inner membrane, and a central core containing the viral genome. This complex architecture is closely associated with the diverse functions of ASFV proteins. Structural proteins contribute to virion assembly and stability, while membrane-associated and non-structural proteins participate in viral entry, intracellular replication, host-cell interactions, and immune modulation.

p72, encoded by the B646L gene, is the major capsid protein of ASFV and one of the most abundant structural proteins in the virion.
